Reengineering the collision coupling and diffusion mode of the A2A-adenosine receptor: palmitoylation in helix 8 relieves confinement
نویسندگان
چکیده
Background: The A2A-receptor engages Gs by restricted collision coupling and lacks a palmitoyl moiety in its C-terminus. Result: Engineering palmitoylated cysteine into the C-terminus relieved restricted collision coupling and resulted in accelerated diffusion of the agonist-liganded A2A-receptor. Conclusion: Restricted collision coupling arises from limits imposed on receptor diffusion. Significance: Agonist induced confinement of the A2A-receptor in a structure consistent with a lipid raft.

Reengineering the Collision Coupling and Diffusion Mode of the A2A-adenosine Receptor
The A(2A)-adenosine receptor undergoes restricted collision coupling with its cognate G protein G(s) and lacks a palmitoylation site at the end of helix 8 in its intracellular C terminus. We explored the hypothesis that there was a causal link between the absence of a palmitoyl moiety and restricted collision coupling by introducing a palmitoylation site. The resulting mutant A(2A)-R309C recept...

Objective(s): For determining the mechanism of anti-asthmatic effect of thymoquinone, this investigation evaluated the effect of thymoquinone in the presence of selective A2A and A2B adenosine receptor antagonists (ZM241385 and MRS1706, respectively). Materials and Methods: Seventy guinea pigs were randomly divided to 7 groups; control (C), sensitized with ovalbumin (S), sensitized groups pretr...

Purpose: MDMA is a synthetic drug that is originated from Amphetamine. It is used by some people. This material induces some chemical changes in serotonergic and dopaminergic neurons in middle brain. Materials and Methods: In this study, we used 49 Rats Sprague-Dawley, 200-250 gr. There were 7 groups and 7 Rats in each group. Rats received MDMA in the first group, in the second group CGS (A2A r...

Background :Adenosine is an endogenous purine nucleoside that has a neuromodulatory role in the central nervous system. The amphetamine derivative (±)-3,4-methylenedioxymethamphetamine (MDMA or ecstasy) is a synthetic amphetamine analogue used recreationally to obtain an enhanced affiliated emotional response. MDMA is a potent monoaminergic neurotoxin with the potential of damage to brain neu...
متن کاملCEREBRAL BLOOD FLOW REGULATION IN ANESTHETIZED MORPHINE DEPENDENT RATS: THE ROLE OF THE ADENOSINE SYSTEM
Adenosine has many of the characteristics of a regulator of cerebral blood flow and adenosine receptors change in morphine dependency. In this study the changes in adenosine receptors' responsiveness of pial vessels in the hind limb area of the sensory cortex were evaluated in morphine dependent rats (MDR) using the laser Doppler flowmetry technique. Adult male Sprague Dawley rats (250-350 ...
